How to Use the Nanodrop Spectrophotometer


Nanodrop spectrophotometer is used to measure DNA or protein concentration or purity.

    The Nanodrop machine and computer are switched on. Open the nanodrop program.

    Add water to the point where the liquids are to be added. Click ok on the pop up window. This will initialize the software. Blot dry the surface with a blotting paper.

    Add the control liquid, eg Nuclease free water. Click blank on the software window. This will set the software values , taring these values as zero.
  4. 4
    Blot the surface of the interface again and add DNA. Click measure. The various parameters and OD ratio 360/380 ratio will come. For pure DNA, it should be 1.8. Protein contamination lowers it and RNA contamination elevates it.
